Acyclovir(Zoviraxâ)為非環狀鳥糞嘌呤核苷酸類似物,其被單純性疱疹病毒thymidine kinase轉變成acyclovir monophosphate,接下來細胞內之酵素再加上一個或二個磷酸根使之轉變形成 diphosphate 及triphosphate,而 acyclovir triphosphate即為活性型。Acyclovir triphosphate會干擾單純疱疹病毒的DNA polymerase,因而阻斷病毒的複製,同時增長的 DNA 鏈可能也被病毒的DNA polymerase 介入,因此終止了進一步的增長
