Everolimus 簡單介紹~~~機轉：Everolimus是一抑制mTOR（mammalian Target of Rapamycin）蛋白的口服標靶藥物，藉由抑制mTOR蛋白，進一步抑制腫瘤細胞生長、血管增生及代謝功能，達到減緩腫瘤細胞生長、擴散或凋亡。 mTOR主要功能為細胞增生訊號之傳遞及細胞週期的進行。如圖一所示，mTOR蛋白接收來自上游PI3K/AKT途徑的刺激，當mTOR蛋白活化後，可以帶動下游一系列的細胞訊息傳遞，進一步促使細胞產生分化或血管生成。在正常細胞中，血管增生的機制受到VHL（von Hippel-Lindau）蛋白的調控，而在腫瘤細胞，這樣的調節機制失去了平衡，導致腫瘤細胞不正常的增生及分化不良。PK：高脂肪飲食會降低AUC及Cmax。Everolimus是CYP3A4及P-glycoprotein（...
